Preparation and storage

Assay Principle: This kit was based on enzyme-linked immunosorbent sandwich assay technology. It was precoated with an anti-TNF-alpha antibody on 96-well plates. And biotin-conjugated anti-TNF-alpha antibody was used as detection antibodies. Standards, test samples, and biotin-conjugated detection antibodies were subsequently added to the wells and washed with wash buffer.
HRP-streptavidin was added and unbound conjugates were washed with wash buffer. TMB substrates were used to visualize the HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue product that changed to yellow after adding an acid stop solution. The density of yellow is proportional to the amount of TNF-alpha sample captured on the plate.
